Mane Six bestest friends?

They are evenly friends. No one is more friends with each other, or it wouldn't be Mane 6.

 

As for bestest friends, I would say something like Starlight Glimmer and Trixie. That's a friendship that extends friendships. (Not Mane 6 though)

Fluttershy seems to have the deepest relationship with everyone. Closer than Twilight's relationships, closer than Rainbow Dash's, closer than everyone else. She's just that kind of pony.
