Perhaps a race? Maybe an adventure through the Everfree Forest? Drama: Finding a mate? Perhaps gaining a cutie mark? How about two siblings fighting for the affections of the same pony? Comedy: Perhaps trying to find a job? Maybe dealing with a disease with humorous symptoms? Parasprites? Horror: Zombie ponies? Maybe a night in a haunted castle ruin? Perhaps a stalker is following you? Genre: Steampunk ponies? Sailor ponies (a la Sailor Moon)? Future-Tech ponies? Anime: Demon-Hunting ponies? Mecha ponies?
