Anyone feel like Applejack is like Twilight?

A big organizer she's head of her household. Often times she can overwork herself as a workaholic a lot like Twilight is. She learns similar lessons. She also had Spike as her assistant once. And she also has a big brother. She also appears to be a capable leader.

It would make sense for them to be close friends. I also thought she would turn out the loyal and close friend to the main character-Twilight.

In some ways yes, they are both relatively mature and level headed most of the time though I think Applejack has a bit of an edge over Twilight here as even when Applejack has freaked out it hasn't been nearly as bad as Twilight's freakouts. They have close relationships with their older brothers though they tend to show it a bit more with Twilight though with Applejack it is heavily implied as she is close to her family in general. They both can overwork but in different ways as Twilight tends to do the more mental work and Applejack tends to do more physically challenging "hard labor".

I've considered them to be almost opposites; Applejack is practical and does manual labor, and Twilight is studious.

After reading this I see that they are alike in many ways as well.

Huh, I never noticed it, but you have a point. I think their leadership roles are focused in different areas, though - Applejack is an everyday leader, managing the farm's business and family matters, whereas Twilight seems to rise to the occasion in times of crises and events.

Somewhat? I mean, of the six, they are easily two of the most mature and rational. They're also the best leaders. Note that whenever Twilight is absent or somehow isolated from the others that it is Applejack who takes charge and calls out the orders, giving her that aura of a leader. I like to think of Applejack as Twilight's second in command, or right hand mare. The way I see it, Twilight is a great planner who always has the best ideas, but Applejack is a bit more stable, less panicky, and has the backbone and quick thinking to help put the plan to action.

Twilight's studious nature is similar to that of Applejack's workaholic ways. But one clearly values education and knowledge where the other favors hard labor and domestic duties. Their interests, values, goals, and priorities are not one in the same.

But while they are both similarly warm and caring, they have numerous differences too. For one thing, Twilight is very organized where Applejack is bit more messy. Twilight is a magical master with great mental capabilities but is a bit of a physical slouch, in contrast to Applejack who has great physical prowess but zero magical ability or even the most basic of understanding of it, and while not dumb by any means, is not particularly book smart either. And where Twilight looks to science and known facts for explanations to things, Applejack is far more likely to put to stock in "old pony tales" or the existence of curses and other paranormal/weird phenomena, like the Pinkie sense. Twilight is more of an indoorsy, quiet type of pony who has a bit of a socially awkward side, unlike Applejack who loves the outdoors, is very charming and socially adept, but boasts a rough and tumble, sometimes aggressive and confrontational demeanor.

Actually yes, I see what you mean with the similarities. Applejack compared to Twilight also possesses a lot of know-how and is smart, but it's a more worldly sort, born of a more personal, applied experience than Twilight's abstract, 'scientific' way of learning. Both are educated, just in vastly different systems (which both work), so I would argue that there are similarities to be found even there, it's just differs in how the core values are expressed.

I think (at least from what I have seen so far) that Twilight inhabits a curious space between Applejack and Fluttershy; smart with relatively strong leadership qualities like Applejack, but a bit more insecure and concerned about how other ponies judge her, especially Princess Celestia.

Another similarity is that they hate failure and take it hard. When Applejack didn't win a blue ribbon or bits she didn't send a message back to the others until she could get the bits she also has to prove she can buck all those apples.

Twilight also dreads failure and takes it hard when she was Celestia's student and even now.

It thought about this comparison, to the point that, in Twi's absence, AJ would make the best leader out of the Mane6. She's quick to take initiative, organized, hardworker, inteligent, and to a point, charismatic. Problem AJ has compared to Twi, is that she's awful outside her element (high society), while Twi adapts better
