I have over 135 hours on the original Isaac.

 

 And Edmund has estimated that for most players, you'll need somewhere around 500 hours to 100% Rebirth.

However, Isaac randomly generates everything. A run from start to finish takes about 40 minutes to an hour and a half if you don't die. But every single time you play, you get different rooms, enemies, bosses, and items. You also unlock more characters, and other various things the more you play, adding even more variety.

But seriously. Rebirth is coming out in less than two months, it has more than double the content of the original, it's going to be on the Vita so I can literally play it almost anywhere, and there's a full-sized DLC that's probably going to come out eventually...

 

 I'm never going to escape the basement this time...
