The weird thing is that she treats him a lot like her family. But calls him a assistent however in the same time Spike can grabs Twilight and even ride her that she is not going to care.

It's not weird that she treats him like family, because she IS his family.

 

I am not sure about the  "mommy" part though, because it's been shown no one, not even Spike, knows who his mother is....I think that was just funny gag bit put it for the lolz.

Considering that Spike was taken as an egg and knew no other dragons until Crackle and the gang, Twilight would be his primary parental figure in life. So yep, it's a guardian-child relationship.

That's not even to mention that it was Twilight's magic that caused Spike to be born. I think she's more than just a foster parent, she's kind of a legit parent to Spike. His egg was born with the specific purpose of being born through magic as a test for young Unicorns.

I never saw a motherly relationship between Spike and Twilight, I always saw a sibling one.

It seems like a combination of a motherly and sisterly relationship with some elements of a friendship, Twilight probably got some help raising Spike but she pretty much did still raise him. Since Spike dosen't know any other family Twilight pretty much is his family.

A good indicate is the episode Winter Wrap Up: Spike specify says he doesn't consider Twilight his mother and later calls her "sister".

 

Also, Spike does too much for Twilight for any parental relationship. This includes feeding and giving her life advice (ex: EG).

 

 

Though I can't say I care about what type of relationship they have after seeing the things allowed to happen to Spike in season 4.

I always sort of looked at it as an older sister, younger brother relationship. Although, I feel like it changes at times. Sometimes it's like a Mother and Son relationship, other times it even looks like Mistress and Servant.

 

So I think the relationship isn't always consistent. But it's still usually very cute.
