1. In The Last Roundup, the humor surrounding Derpy worked and was funny because she wasn't the joke, but because of the exchange between Dash and Derpy bounced off each other.
2. In Canterlot Boutique, the joke with the fat mare doesn't work because the mare herself IS the joke. It's a visual gag unrelated to the entire story, conflict, and resolution.
