There's bound to be some jokes in the movie about the cultural differences between the pony world and the human world. We've already seen Twilight eating the apple in the trailer. Here are a few jokes that I think will be in the movie.

 

Twilight will use the expression "everypony" and everyone will be confused by it.

 

Applejack might not be an apple farmer in the film, but at some point she will have to get something down off of a high shelf or something and she will just kick the wall to get it to fall down.

 

When ponies go into the human world they turn into humans, when dragons go in they turn into dogs. So what will the human world version of Winona look like? Maybe she'll be a dragon XD but she'll just be one of the little lizard kind like we have here on Earth.

It's hard to predict gags IMHO since they stem from the individual scenes and situations that they occur in.  Guessing gags beyond what has been in the trailer is pretty rough since we don't exactly know what scenes will be happening beyond the premise.  We know they'll be jokes around Twilight still thinking she's a pony, Spike snarking left and right about everything like he always does, and of course they'll be little fandom nods here and there (Scootaloo was dancing like a chicken for crying out loud!).  After that, the laughs could come just about anywhere.

 

I'm not going to predict.  I'm just going to sit back and enjoy the funny as it comes.

It's kind of likely there might be a few of our favourite background ponies in there, we've already seen Vinyl Scratch, who's to say that we won't be seeing any of the others, I, for one, predict that Octavia shall be in the school orchestra. Something else that's also been on my mind, what about Diamond Tiara and Silver Spoon? Will they be harassing the human equivalent of the CMC? And, leading on from that, what will be the human equivalent of a Cutie Mark, otherwise the CMC would be really boring...

Oh my freaking goodness, those would be awesome. Here are some of my predictions;

 

Pinkie Pie will get arrested/detention for using a party canon in school.

 

Fluttershy will try and be all friendly with some animals and get attacked.

 

Rainbow Dash will try out for track and accidentally do a sonic rainboom. xD Or she'll try and save somebody and jump off of something forgetting she can't fly.

 

Can't think of anything for Twi-Twi and Rarity though. D: And I like your AJ one.
