No. Apparently you have to kill the game's first boss in his level, without killing any other enemy, and you have to do it on a time limit.

 

 But he only spawns when you open the weapon and ammo chests on the level. So you have to do that first. But the levels are randomly generated, so they might spawn behind a wall of scorpions, constantly shooting at you. Making them impossible to access without either killing them, or dying yourself.

 

 But apparently this is wrong. Since some people have gotten the secret without doing this...

That seems stupidly hard... How are you supposed to get to him if you can't kill the things guarding him? I guess there is a way, but yeah, doesn't sound fun.

Somebody just released a video of them getting to it. He killed some of the enemies but still got there...But he has no idea how...
