I can see Spike getting a G5 incarnation due to he popped up in previous Gens(except for one of them I think).
I can see Princess Luna/Nightmare Moon coming in due to how she was really popular and how Nightmare Moon was the antagonist they kicked the show off with her/Luna's story and being the first antagonist they fought.

Discord definitely is a strong candidate for a return. He's very dynamic and unique and adds much flavor and a different perspective to the show. He also makes for an interesting plot device and counterbalances The Mane 6, Celestia and status quo in general, whether it's as a villain or ally.

I may just also be a Q and John De Lancie fan, but demigod like characters like Discord for me can really amp up a show when well executed.

I think it's safe to say that all of the mane 6 will probably reappear, just due to popularity, but with it is other characters are certain. But I can see Hasbro bringing in chracters like Luna or Sunset, just to please the fans.

I'm kind of in two minds. On one side, I would prefer it if it continued in the same world as before, since I've become so emotionally invested in these characters and I would really love to see them return. Maybe a sequel series set a few hundred years in the future might be nice (sure, most of the characters from FIM would be dead), but we can still see the impact that they each had on Equestria, through books and paintings.

On the other hand, a new start would also be nice. It allows us to explore a totally different world. with new characters. If done well, it could be as good or even better than FIM.

I think next generation won't be related to FiM because it would be next season, spin-off, but not G5. However, I don't think they will abandon FiM, they'll at least put easter eggs. Or maybe it will be other place or time period in pony world Fallout Equestria.

It gave an idea that setting might be a far future Equestria where ponies developed technologies to a human level and mane 6 are just another page of history book
