Imagine if you will one of the main six has now turned evil.  your called in to pick up her element of harmony (or maybe even with a new one that better suits you).

I'd say flutter shy.  Imagine having totally defeating her.
Me
"What do you have to say for yourself young lady."
Fluter shy

*all quite and meak.* "I'm sorry can i go now?"

Me
"Daw.....well alright."
Flutter shy "

Yay i'm gonna go back to ruling equistra."
Me

"GOD damn it that's the fifth time she's done that!"

it has to be pinkie TBH, I mean just look at smile HD!

How can anyone stop that??

She would be dropping smile bombs everywhere!!

But I think overall they would be defeated, I mean the power of friendship trumps all right? so long as there where still friends in equestria!

Twilight Sparkle, especially considering her advanced skill with magic. She has numerous spells that could easily put down any opponent and she continues to learn more because of her insatiable hunger for knowledge. She could obliterate you before you even make your first move. Quite a deadly opponent indeed.

Twilight would be the most difficult to stop due to her being the one of the most powerful magic users in the history of Equestria and her fall to darkness may come with a bit of a power boost. King Sombra is a prime example of just what can happen to a unicorn if they are advanced enough in magic and fall to the darkness. It would still take a bit of time for Twilight to become that powerful but would still be very difficult to stop because of this.

All of them have enough abilities to be very dangerous if any one of them became evil.  With that said, the toughest opponent would be Twilight Sparkle, without a doubt.  Her advanced magic would make her too much to handle.

The second hardest pony to apprehend would be Pinkie because of her cartoonish abilities.

I doubt someone else could come in and assume the element of the pony that became corrupted.  Twilight tried giving Dash's necklace to Spike and that didn't work.  Those just serve as conduits for harnessing the elements.  I believe real power lies within the ponies themselves.

Geez maybe twi or rainbow dash but then again maybe applejack you never know we can all agree rarity would be easiest to defeat

It'd definitely be Twilight. The show builds up the idea that Twilight has unfathomable amounts of magical abilities. Without a good soul keeping her in check, this magic could wreak unbelievable havoc on all of Ponyville. She could easily give Equestria a run for its money if she had all her will set on destroying or overtaking it. She could easily take down her friends, Ponyville, and she could probably get pretty far through all of Equestria. If Friendship wouldn't be the answer to returning her to normal, it would probably take the Princesses to really stop her.

I'd say definetly Twilight Sparkle, even when she's using her magic for benevolent purposes she's incredibly powerful. Now if she went evil and her moral compass got flipped she'd be probably unstoppable, magic being the most powerful anything in all Equestria and Twilight herself devoting herself day and night to studying magic and being the best of the best at it... because apparently she's suppose to become the next Starswirl The Bearded. And after writing this paragraph I myself have a new found respect for Twilight and her abilities.

Definitely Dashie. I can't possibly catch up to her if she's flying so it would be partially impossible for me to stop her immediately.

Hmm I would say Twilight as the rest could be dealt with quite easily.

People overestimate Pinkie's nature, Applejack would be easy, Fluttershy & Rarity would be a cinch and Rainbow Dash would provide small difficulty.

Twilight being an alicorn would be difficult to beat because of her magic. She knows a variety which include mind wiping and transfiguration spells which we have no way of countering so she'd be very difficult to beat.

I'm gonna go with Pinkie Pie on this one.  Twilight's power is quite formidable, but Pinkie's is inexplicable.  When Rainbow Dash tries to escape Pinkie, Pinkie is already waiting for her at her destination.  She's not merely faster than RD, she's non-local.  In the episode where Fluttershy becomes a model (I don't recall the title offhand), Pinkie Pie exhibits a kind of omniscience and omnipresence.  Even though the other characters can look around the room and know that Pinkie is somewhere else, they also know that she knows what they're saying and can appear at will ("Forrrreeeeverrrrrr!").  IOW, Pinkie could make a "stealth roll" for an assassination attack--and escape afterward--with surpassing ease.  In contrast, Twilight can only project power where she is physically present.  She can only see and perceive through her own senses and scientific instruments.  Her speed is mediocre, and her teleportation power is limited to a relatively short range.

The show presents us with plentiful examples of Twilight's combat ability in action, so we have a pretty good idea of where her limits are.  We don't know where Pinkie Pie's limits are, or even if she has any.  Example: in The Crystal Empire, Twilight is trapped by a growing wall of crystal and has to send Spike to bring the Crystal Heart.  Would Sombra's trap have even slowed Pinkie down, were she in Twilight's place?  Or would she have just emerged from out of a potted plant with the Heart, leaving Sombra's trap empty?

Feeling Pinkie Keen presents us with the closest thing we have to Twilight and Pinkie being in combat with one another, so it offers us our best canonical demonstration of relative power.  Twilight gets punked again, and again, and again.  We see her with her limbs in casts and in traction as a result.  Then, she's driven to a complete psychological breakdown.  Twilight doesn't take a shellacking like that from any of the show's major villains.  Yet, as far as we can see, Pinkie isn't even upset with her, much less trying to defeat her.  We are simply shown that if you mess with Pinkie Pie, the universe becomes your enemy.  This is capped off with a demonstration of Pinkie's sense seemingly yanking Princess Celestia right out of the sky.

Though the Pinkie Sense probably doesn't cause the events it detects, and it is not under Pinkie's conscious control, it indicates that Pinkie Pie transcends mere personal power.  It's a "Pinkie" Sense, thus apparently metaphysically unique to her (unlike, say, "being psychic").  It appears to be infallible, and operate on a basis of reverse causality (events in the relative future cause Pinkie to twitch in their relative past).  In Equestria Girls (non-canonical source, but still...), Pinkie and her counterpart demonstrate complete knowledge of parallel universes they have not visited.  Keep in mind that when Celestia sent Twilight through the mirror, she said that Twilight would soon know more about the world on the other side that even she did.  Pinkie knew more than Celestia without having to go through the Mirror.  Combine this and the Pinkie Sense with her other abilities: Pinkie Pie transcends space and time.

When Twilight challenges Pinkie, she is punished severely, with Pinkie herself not needing to lift a hoof, or even think about lifting a hoof.  In a nutshell: Pinkie Pie kicks Twilight's ass repeatedly, in the most hardcore fashion (harder than all of the show's villains put together), on such a "meta" level that Pinkie doesn't even have to want to fight, in order to win.

Twilight, by far. I believe it was Discord in Season 2 Episode 1 who said that Twilight had the most powerful Element of Harmony, Magic. If you think about it, Twilight has an insane amount of Magic, just think of all the chaos she could do to the world if she turned evil. Also, the other elements wouldn't even work right without Magic would they? You know... "and Magic makes it all complete." I'm sure if she did go bad then she'd start studying black magic, which would probably rip Equestria a new one. Opinions! Opinions everywhere! :D
