Nightmare Moon's laugh was the best for me. If for nothing else, then it was the classic villain laugh that you can't not love.

 

Or else it would be Twilight's crazed laughter during Lesson Zero (I think?).

One of the things that has always drawn me to Rainbow is her voice, and her laugh, in particular, has always been endearing and infectious.

 

I mean...  She just can't help herself. xD  Rainbow does try, but her mirth can not be suppressed.
