Twilight Sparkle would immediately become the sole Princess of Equestria, transforming the country into an absolute principality. However, the tasks of raising the sun and moon, running the country, going to formal events, and receiving foreign dignitaries would become incredibly taxing on her, as she lacks the same levels of composure, knowledge, and experience that Princess Celestia has.

 

I'd would wager a guess and assume that Twilight would at first try to do everything Princess Celestia could, probably out of respect for her late mentor and to ensure full continuity of government. Eventually, I could see Princess Twilight shifting most of the day-to-day administration of the country to a lower body or organization, regardless of whether it's the nobility, a separate civil bureaucracy, or an elected parliamentary body, while she continues to raise and lower the sun and moon and receive foreign dignitaries and nobles.

The answer is simple:

She would basically take the role everybody used to have.

The only problems she would basically have would be stabilizing the Crystal Heart and getting enough magic to control the Sun and the moon, assuming it happened in present time.

If I was Twilight, and the last Alicorn, I would delegate the rising and setting of celestial bodies to a team of Unicorns, like in olden pony times. That would leave her free to do the actual ruling. I would also appoint a governor general to hold office in the Crystal Empire. If Shining Armor survived whatever killed his wife, he would be the logical choice.

I don't know about cadence but I've always been of the opinion that Celestia was getting twilight ready to take over because both she and Luna are loosening their power and thus their immortality.

We have seen from the Tierek episode that Twilight can handle the power of the 3 other alicorns, since she's the princess of Friendship she would probably make it first priority to calm everyone down. Then assuming what killed Celestia, Luna and Cadence didn't kill the royal guards, Twilight may select a few to help her move the Sun and the Moon (because we saw how well that worked alone!) Then she would go comfort Shining Armor and help him take control of the Crystal Empire. If this all happens I will be extremely sad because Celestia is best princess!

Nationwide grieving of course. Then Twilight would have to assume command of central Equestria in place of Celestia and Luna. She would likely assemble a dedicated team of Unicorns to raise and lower the sun and moon like it was in days of old. Shining Armor will of course, continue ruling the Crystal Empire by himself. It would be a very demoralizing era in Equestrian history.
