Trixie and the Alicorn Amulet

Honestly, given the way actual myths and tales about similar magic artifacts go; the amulet's magic boosting will still have a cost. Very likely, the amulet would consume Trixie in some way and take control.

For a well-known example, the Ringwraiths from the Hobbit/Lord of the Rings were originally humans who fell under the influence of nine magic rings bound to the One Ring. The power in those nine rings boosted them, but reduced them to undead monsters. This is pretty typical in myth, so I think it would be reasonable to assume something similar would happen to Trixie eventually (and for that matter Rarity when she fell under the influence of dark magic from that book Spike found.)

Celestia would step in of course. She's not known for showing up for disasters in Ponyville all that often, but I think that's because Twilight's there, and in order for her to grow and become independent, she has to learn how to solve problems on her own. If she couldn't manage the well being and safety of a small town, then she wouldn't be fit as an additional ruler in Equestria. The only time Celestia has stepped in so far is when Twilight was the source of the problem.

Trixie might not exactly live up to her self imposed title, but she is more skilled than your average unicorn. Still, even with the Alicorn Amulet, I think she'd exhaust herself trying to hold out against a true Alicorn and the Royal Guard. Before long she'd be another stone peice in the Canterlot gardens.

Celestia, Luna and the Royal Guard would eventually defeat her. Alternatively the EoH would also beat her. Amulet Trixie is powerful yes, probably as strong as Celestia or close enough; but there are things in MLP that can easily defeat alicorns so....
