i'm gonna go out on a limb and say this comic is about luna.

i would think coffee would be like luna's Cryptonite, being the alicorn of the night, and all things sleep,

 

is that what the issue is about?

anyway, i think your assumption would be pretty spot on. tough luna is often pictured as the young&wild kinda type by the fandom, so i guess that could be a reason too.

If she is awake the normal 15 to 18 hours of the time, drinks it at the intervals normal coffee drinkers do and avoids caffine 3-5 hours before she sleeps why would it effect her worse then someone who stays awake the day? Why would coffee be her cryptonite anymore than anypony else.

 

To much coffee however can be a problem... Those eyes.

I think it also goes along with the whole "gamer Luna" meme. I guess gamers/computer geeks drink a lot of coffee and plus she's the Princess of the night, she needs to stay awake.

It could be. But, there are all kinds of covers with different characters on them and they don't either show up or it's just for fun.

Now I know there are dozens of comic cover where characters are dressed up like superheroes. But this cover does not mean it's about the Cake's or even the twins. It's just a fun cover. So this could be a cover saying a Luna comic is on the way or it could just be a fun cover.
