Only if you view G5 as a truly direct sequel to G4's universe and assume that they all died, rather than something happening to them that effected them via an event that caused the world of G4 to change both physically (from 2d to 3d) and figuratively. For all we know, they (or at the very least, the mane six) could be in stasis, which would still leave them still alive.

Well, we know Twi is not in G5 at all - confirmed by Tara Strong in a livestream.

But yes, it is possible that G4 never even existed in G5, and that the legends of the Mane 6 there are purely fictional (and just by coincidence look like the G4 ponies)

It seems unlikely though.
