It all depends on how they would handle the thing. Seriously, take most of the aspects of the show and put them in human form and they would still work to a degree.

I like humanized ponies when done well. They have to reflect the personality, mannerisms, and whatnot of the characters. Slapping on the color patterns of ponies doesn't cut it.

 

The same goes with any franchise. Even ones like Pokemon where the characters don't behave like humans. In the case of Pokemon, this is a really well done example where the physical attributes (including weight, height, mood - more than just colors or distinct features) of the Pokemon translate to the humanizations, whereas a bad one would be like a generic dude with no distinct physical attributes with Venusaur-colored clothes and a flower on his head.

They should do what they did with The Powerpuff Girls and ship it off to Japan for an anime retooling.

 

They could call the anime version something ultra cool like All-Purpose Cultural Pony Girl Twilight Sparkle, but considering the state of the business these days, I'm guessing they would probably go with something gallingly orthodox like Pony Power Z.

 

The Mane Six would be a super sentai of high-tech, katana-wielding ninja warriors who fight crime. They would take on the big bosses by assembling into a giant ponybot called Princess Celestia 9000, who would confront villains in downtown Tokyo and wantonly destroy half the city as they fight the villain of the week.

 

They would all, of course, be high school students who would keep their true identities secret, would have a cute little moe-ball cat- or rodent-like pet with magical powers, would have exaggerated traits (Twilight would be even brainier and nerdier, Rarity stuck up and more fashion-obsessed, Rainbow a bullying jock, etc.) and would have at least one beach and/or bath house episode per season. And possibly more.

 

They would, of course, be fully human, except for their pony ears and pony tails, which they would either hide or cleverly integrate with their obligatory sailor-style school uniforms and "civilian clothes". Oh yes, and Spike would be a little green alien with an insatiable appetite for wagashi.

 

It would probably flop in Japan, like most such shows do, but they could make it up shipping cheap DVDs and doujins to America and selling them for ridiculous prices to drooling, OCD-ravaged Yankee otaku.

 

It's the perfect plan. Hasbro should hire me to make this happen.

I dislike humanizated ponies for some reason, not sure exactly what is it. I wouldn't like it, so for me, it would be just another part of fanwork.

I like the humanization of ponies so long as it is done correctly. That is to say, they need to be fully human; not a mix or cross between pony and human. The only exception to that is allowing horns and wings just to keep it easier to understand the show and characters. These two drawings are my favorites for humanization:
