It is Twilight Sparkle without a doubt, she has gone from an anti social mare with her nose always in a book to still being somewhat introverted but starting to have some very close friendships. I have doubts about her alicorn ascension and was very much opposed to it for various reasons but we will have to wait and see what happens and hope that we don't get another screw up like Magical Mystery Cure. Rainbow Dash comes in a fairly close second though as while she is still a bit reckless and cocky she has matured the most out of the mane 6 as we have seen in Wonderbolt Academy and is starting to slowly become less reluctant to express her feelings as we saw in Sleepless in Ponyville.

Twilight has of course gotten the most development, since she is the main character like 40% of the time.

Dash is probably second then, especially with her skills as a flier I feel.

Pinkie has gotten some good development with the Pinkie Sense thing and her ordeal with Cranky but not much has been done with those since.

Rarity has had some development in her career I suppose, and social status, and of course with her sister.

Fluttershy I feel has some decent development going on with the 'being assertive' thing, but it's really back and forth I've noticed, I think she needs more lessons. ^_^

Applejack has gotten some development in working too hard and perhaps in keeping secrets, but she could use a bit more.

I'm gonna go with least being fluter shy, (even though she's my favorite)

she lapses back to her old self.
I'd be hard pressed for you to point to me how season 3 flutter shy is different than season 1.

I could say the same of apple jack, but she really did grow from her lessons back than it would seem.

As for most?  I'd like not to say twilight just so we could have a discussion. i mean it's not much of a thread if we're all in agreement.

kinda hard to deny the one with the spot light grew the most.  if she didn't it would be a pretty bad show.

Twilight Sparkle and Rainbow Dash probably have the most character development, when it came to the episodes that centered around them and how much they have grown since the show began. Rarity has some character development when it comes to being with her sister, and her social status. Fluttershy has a decent level of character development, when it comes to her personality and Applejack's character development comes in small doses, when it comes to her family and her own issues.

I think season 4 will offer a lot of defining character development milestones for the rest of the mane 6. I think from then on we could get an idea besides Twilight sparkle who will have the most, though it would be great if the rest of the main 6 had something equally as important, though it can not be bigger than becoming an alicorn princess but each will be epic none the less.

Applejack. She's the only character that approaches the three-dimensional status, having a near complete family, a job, a realistic personality, a "good" accent, and she's so great they had to turn her into a background character because people want to see her more.

After AJ probably Pinkie, since she does have a family and a job, but she's not far from being one-dimensional thanks to that silly toon personality of hers.

I also consider Fluttershy, but she doesn't exactly have a job nor a family yet. She does seem realistic and relatable to most bronies who are shy to meet others.

Old joke is old (though I admit I think you said that with a healthy dose of sarcasm).

I already spit out nearly 1600 words of pro Rarity propaganda earlier so I'll agree that Applejack probably is the most realistically fleshed out character of the Mane Six. Twilight has had some major moments the the Season 4 opener and finale. Dash, Rarity, and Fluttershy all require exploration of some degree but should you endevour to do it ... you can find your self looking at as real a character as any in fiction.

Pinkie defies both simple and complex explanation ... she is Pinkie Pie and that is more than enough for me. :)
