Well, I think Twilight would not be phased, considering her trip to the human world.  I feel most ponyfolk would find humans to just be odd creatures they didn't know about, considering they live in a world of mythical creatures.

 

For humans, however, I think they would not take it well.  They came from the human world from MLP, if not our world, where magic and talking animals don't exist for them, so it might be too much of a cultural/reality shock.  They would probably think they drank some funny water.

Probably the same way we would respond to aliens visiting earth, but a little less dramatic because they are used to strange creatures.

 

If that would happen, they should be careful. Humans have a tendency to start wars and destroy ecosystems.

If humans wandered into Equestria, how do you imagine the ponyfolk will respond. The humans are not guised as ponies, and no, they sadlydo not find anywhere to hide. What do you think?

 

If we wandered into Equestria, it would most likely be instantly ruined.

Among the humans a type of Xenology would pop up based around studying the cultures and biology of Equestria. The government or UN would probably regulate the pathway/portal to Equestria, allowing only for highly trained specialists and scientists to venture there over very short periods of time, with the hope of bringing as little cultural contamination into the Equestrian world as possible. Eventually the ponies get curious, catching brief glimpses of foreign, computer-based technology held by the scientists, and picking up on the foreign words used by the human scientists such as "hand" and "phone" and "computer". The ponies will insist to be allowed to learn more about the humans and their world and technology, claiming it unfair how the humans can study them, but the scientists are unable to share anything due to the government regulations. Eventually Twilight comes to them, showing her knowledge of the human world which appears to the government as heavy cultural contamination, which gets the scientists in very big trouble. Twilight and the ponies feel bad about this, but two of the scientists end up escaping prosecution by fleeing back into Equestria. The humans then launch a huge military invasion of Equestria, with the possibility of bringing with it the first Xenocide of human history, for fear of the ponies invading the human world and using their technology against them along with magic (this invasion ironically brings about massive international cooperation in the human world). The scientists that escaped, however, decide that humans suck and they want to work together with the Princesses and all of Equestria to defeat the humans and bring about peace between the worlds. Even the dragons, the griffons, and the yaks of Yakyakistan help out, and many people in the human world begin to protest and fight for Equestria. And so begins the first Interdimensional War of humans and ponies.

 

The Equestrian side wins the war after many years of fighting. Humans are devastated by the loss, but the ponies help them rebuild and eventually teach all of mankind the friendship of magic, ushering in a new age of world peace.

 

The End.
