I think they might all be bi. The reason is because most of Equestria seems to be populated by females. If they're all bi, then it's easier to find love. If they're hetero, it may be hard because the minority of the population are male.

Twilight: Heterosexual. Generally she seems uninterested in sexuality, putting more emphasis on her studies than on romance. But she displays a definite romantic tension with Flash Sentry.

Rarity: Heterosexual. Clear evidence of attraction to the opposite sex on several occasions.

Rainbow Dash: Homosexual. Every time she's with Fluttershy there seems to be a desire to protect that goes beyond mere friendship.

I'm late to this, but my headcanons are
 

Twilight: Bisexual

RD: Lesbian

As you can see, I don't see any of them as straight, but I have a reason for this.

A large majority of the pony population is biologically female, so it's only logical that most mares would be bisexual at the very least. :3

However I would like to think that literally every single character in the show is not except for Rainbow Dash.

I would find it funny if she was the sole straight character in the entire show.

I am think most of them would be heterosexual, with the exception of Rainbow Dash and Fluttershy, which I will explain in a minute.

 

Even though the show wouldn't say directly, definitely Twilight and Rarity are heterosexual, due to being shown to like male characters, like Trenderhoof and Flash Sentry (don't kill me please). It may be possible for them to be bisexual, but I don't think I have seen enough to suggest that.

 

For Pinkie Pie and Applejack, there hasn't been enough shown, so it is difficult to tell. I do think they are probably heterosexual. There haven't been any "close" interactions with the others, as far as I can tell.

 

Last is Flutter and Rainbow. I am not trying to stereotype Rainbow Dash, but she seems to be very close to Fluttershy. Not just friends, but it seems like she likes her, and same with Fluttershy. They like to be together, and likes to be close to her. I think they are homosexual or bisexual due to that, though I decided on both being bisexual. Mostly just a preference.

These kinds of topics are always fun.

 

Statistically speaking, odds are that no more than one of them is anything but hetero, and probably none.  I think the percentage of homosexuality occurrence in any mammalian population is about 2%, or 1 in 50, so 1 in 6 is a long shot.  And I think bi is even less common.

 

BUT WHERE'S THE FUN IN THAT?!

 

• Rainbow:  Lesbian.  Honestly, I'm not basing this on stereotypes or color schemes.  I've just always imagined her that way.  She seems like she would prefer feminine beauty in her mate.
• AJ:  Also lesbian, pretty much for the same reason.  And I'm a fan of AppleDash.
• Twi:  Bi.

• Rarity:  Hetero, since that's canon, as she was attracted to Blueblood and Tren.  Technically, she could be bi, but I just don't imagine her that way.

• Fluttershy:  Hetero.  A lot people seem to imagine her as hetero, but not being interested in a relationship.  But I don't know--I think she may surprise you.  But I'm sure she'd want somepony very quiet, sweet, and gentle.

• Pinkie: Definitely pansexual, although I've never really understood the difference between bi and pan.  Bi means both, pan means all.  In this context, pan should mean both as well, because there's only two sexes, right?  Unless I'm very badly mistaken.  Gender is a fluid spectrum, but there's only two sexes.  I mean, there's variations--anomalies, combinations of the two, lots of things can occur, but none of them are a third sex, so...I dunno.  I guess pan just accounts for any of those kinds of variations, as opposed to just average masculine male and feminine female?  I dunno.  Anyway, Pinkie would be pan, for sure.  I could totally see her going off on some high pitched, energetic, screaming spiel about how all ponies are equally beautiful and she couldn't imagine not finding any of them attractive.  (Methinks I overthink these things....)

In my headcannon, Twi is straight, Pinkie is pansexual, Fluttershy is ace, AJ is bi, same for Dashie and Rares is straight

Polysexual. Her attraction to Flash Sentry aside (depending on whether it counts as canon outside EQG), I would like to imagine her as having some sort of attraction for any sort of gender for experimental purposes.

Heterosexual. It's pretty evident in her attractions to Prince Blueblood and Trenderhoof, and quite possibly even Spike.

Homosexual. Nothing do with appearance or personalities, but there have been moments where she might have an attraction towards those of the same sex. If there's any sort of hint of attraction towards Soarin, she might at lease be bi.

Asexual. There is a possibility that she might not have an attraction towards any gender.

Bisexual. Though she probably would like to be in a relationship, she's just too shy to make any attempts at doing so.

Pansexual. Due to her hyper personality, I see her as having an attraction towards anything. Not just other ponies, but other creatures and even objects.
