First of all, make sure I can get back home. If so, bring only the people I trust with me. If I was told I couldn't go home, I'd would find some way to close the portal with me outside to prevent the portal from falling into the wrong hands/hooves/whatever. Our world may be more cruel than Equestria, but it's home!

Hahah. For one thing I'd do a lot of phone calls and sending dozens of PMs all over the forums, then I'll try to facilitate eager bronies to enter the portal. I'd watch the entire thing from the side and be happy about it, though I won't enter it myself.

I would probably get the Ponygames Guild and any of my other brony friends we would enter the portal and I would block off the path so no one else finds it, because then Equestria would get overrun by humans and if lets say the U.S. Government (or any government) discovers it they would send troops and probably try to make it there territory peacefully or by force, and if I decide to let other humans in then I would put myself into a great position of power that would make me the lord of all Bronies..... but I'd rather there world not get overran by humans

 

but if I were to of invite everyone in the fandom through the portal and choose to become the Lord of all Bronies we probably would make are own town that would be known as New Jamestown (if you don't get the reference Jamestown was America's first English colony) and Equestria is a parallel world of America so it only sounds right to have the first human town be named after that

 

but that's what I think I would probably go with the first option because I want to avoid a few pretty specific things I know will happen if I choose the second one.........

Depending on what type of portal it is, that may not actually destroy it. It is certain, however, that massive amounts of radiation will leak through the portal, whether the portal itself is destroyed or not. Although now that I think about it, that would be a very good way to make fallout Equestria cannon...
