I imagine this scenario also implies that we're ponies in Equestria. In which case, if Twilight just up and kissed me, I'd wonder "what?" then I'd wonder "why?" then I'd take her out regardless of her answers. If you know what I mean.

Well, my reaction then would be to shove and go GET OFF OF ME, WHAT ARE YOU DOING!? and go see a doctor to make sure I have no diseases- or that would be the case if I was still a human, and that was a pony that just jumped me.  But, if the situation was as you are thinking it should be while you ask this question to me, with both I and said female acquaintance as ponies, then if the setting is right, the moment as well, and the correct social istory is applied, then I would go I KNEW IT!

I would probably be in complete shock that it's even happening and honestly I would probably start having a panic attack or something lol. If I wasn't aware I was dreaming, and Twilight kissed me, I'm pretty sure I'd be blushing and just standing there unbelievable shocked. I'd probably question why she did that, but then I'd probably just go to hugging her with a big smile on my face.

Tough question...Considering I don't have one favorite pony, but more like three. They are Rarity, Fluttershy and then Princess Luna, which are much different then eachother.

 

If it was Rarity I'd act calm at first, return the kiss back, then be marvelled at the fact it's Rarity who kissed a commoner styled stallion such as me.

 

If it was Fluttershy, I'd return it back only to pull away soon after and be shocked that such a shy and quiet little mare would do that in the first place.

 

If it was Princess Luna...I'd be worried at first about the intention of it, but return the kiss back and follow her lead with it due to her past as Nightmare Moon.

 

So....yea. That pretty much sums up what I'd do with each of my favorites.

As a pony: If Derpy kissed me I'd defo kiss back! Even run my hooves through her mane, check out her cutie mark with the other hoof,  I'd offer her a muffin, then I'd most likely flip my tail over my back and prance off like a colt in spring, and worry later about what Dr Whooves might have to say! 

 

As a human: I'd also kiss back, wonder how I became a cartoon, Wonder who spiked my drink, then after all the action died down, (Since outside of the cartoon world, action would be more likely to happen)  and after Derpy went back to ponyville in the Tardis, I'd ring my shrink and schedule and emergency appointment!
